The Gram stain method was developed in 1884 by the Danish bacteriologist Hans Christian Gram. It is one of the most useful staining procedures because it divides bacteria into two large groups, gram-positive and gram-negative. Structural differences in bacterium cell walls affect the retention or escape of a combinaion of stains and washes. The gram-positive cells retain the crystal violet color and the gram-negative cells do not retain the color until stained with a red dye such as saffranin and remain pink at the end of the procedure.
